Flies were washed (five times for 3 min each) with 0.15 M sodium cacodylate buffer pH 7.4 containing 2 mM CaCl 2 on ice, postfixed with 1% osmium tetroxide in 0.15 M sodium cacodylate buffer pH 7.4 for 1 h on ice, washed (five times for 3 min each) with ddH 2 O on ice and dehydrated in an ethanol series (20%, 50%, 70%, 90% and 100%) for 5 min each on ice, then 100% dry ethanol at room temperature twice for 5 min each, then 1:1 v/v dry ethanol and dry acetone for 5 min at room temperature and finally dry acetone for 5 min at room temperature
